Methodology. Each of the 147 markers is an allele that the high-coverage Denisovan genome carries, that none of the
three high-coverage Neanderthal genomes (Altai, Vindija, Chagyrskaya) carries, that is essentially absent in African populations, and that is clearly
more common in Papuans than in Europeans. That last contrast is what makes the test specific: Europeans and Papuans share the same out-of-Africa
history and the same Neanderthal ancestry, so what separates them at these positions is Denisovan ancestry. Reference genotypes come from the Allen
Ancient DNA Resource (v66.p1, 1240K), and the scale is anchored on real reference populations. The panel was validated on individuals held out of its
construction: Africans score about -9, Europeans about 2, Native Americans 31, East Asians 35-54, Melanesians 65, Aboriginal Australians 69 and
Papuans 91-100.
